Abstract

An assay using LC-HRIM-MS was developed to quantify gangliosides at the molecular species level in brains from wildtype (WT), D409V heterozygous (HET), and homozygous (HOM) GBA mutant mice which are a model system for Parkinson's disease.

  • Developed a 30 min LC-HRIM-MS method for in depth ganglioside characterization of mouse brain samples for minimal ion suppression and high throughput.
  • Separated isomeric gangliosides in the mobility dimension that were unresolved in the LC dimension.
  • Demonstrated flexibility of HRIM analysis for high throughput detection and quantitation of abundant gangliosides for of  ganglioside profile characterization.
  • Observed differences in ganglioside relative abundances among WT, HET, and HOM D409V GBA mutant mice.
